TMT-Based Quantitative Proteomics Analysis of the Fish-Borne Spoiler Shewanella putrefaciens Subjected to Cold Stress Using LC-MS/MS

چکیده
Shewanella putrefaciens is a specific spoilage bacterium for fish during cold storage. To better understand the molecular mechanisms of stress adaptation S. putrefaciens, tandem mass tag- (TMT-) based quantitative proteomic analysis was performed to detect effects on protein expression profiles in which had been cultivated at 4°C and 30°C, respectively. A total 266670 peptide spectrum matching numbers were quantified proteins after data analysis. Of 2292 quantitatively analyzed, 274 found be differentially expressed (DE) under compared with nonstress control. By integrating results Kyoto Encyclopedia Genes Genomes (KEGG) analyses, 9 common KEGG terms notable cold-responsive proteins. Generally, DE involved carbohydrate, amino acid, fatty acid biosynthesis metabolism significantly upregulated, leading energy conservation survival mode. The related DNA repair, transcription, translation implicating change gene more needed response stress.

The major soluble cytochrome isolated from microaerobically grown cells of Shewanella putrefaciens has been shown to be a novel type of flavocytochrome with fumarate reductase activity. This flavocytochrome, located in the periplasmic fraction of cell extracts, has been purified to homogeneity and shown to contain 4 mol of haem c and 1 mol of non-covalently bound FAD per mol of protein. متن کاملShewanella putrefaciens – a new opportunistic pathogen of freshwater fish
In recent years, Shewanella putrefaciens, commonly known as a halophilic bacteria, has been associated with serious health disorders in freshwater fish. Therefore, it has been described as a new aetiological agent of the disease, named shewanellosis. S. putrefaciens is a heterogeneous group of microorganisms, belonging to the Alteromonadaceae family. Based on different criteria, three biovars a...
متن کاملShewanella putrefaciens infective endocarditis.
Shewanella putrefaciens rarely causes infection in humans. In the last few decades a growing number of cases have been described. The following report outlines the case of a 40-year-old immunocompetent white man with S. putrefaciens infective endocarditis. This is the first known case of infective endocarditis due to an apparently monomicrobial S. putrefaciens infection, and the second known ca...
متن کاملFirst isolation of Shewanella putrefaciens from freshwater fish – a potential new pathogen of fish
Bacterial isolates from common carp (Cyprinus carpio L.) and rainbow trout (Oncorhynchus mykiss) were identified by the API 20E system (bioMérieux). Twenty one isolates were identified as Shewanella putrefaciens. These isolates were tested for selected biochemical and cultural characteristics. Pathogenicity of two isolates (Pt38 and K116) for freshwater fish was assayed. Biochemical characteris...
متن کاملShewanella putrefaciens abscess of the lower extremity.
Shewanella (Pseudomonas) putrefaciens is a ubiquitous organism that has been isolated from many foods, sewage, and both freshwater and salt water (9). Isolation of this organism from human specimens most commonly represents colonization. However, case reports describing this organism as a human pathogen exist in the literature. Descriptions of localized infections such as cellulitis or abscesse...
